le site de lyon concentre un important réseau d'institutions dédiées à la recherche sur le cancer, disposant d’une visibilité scientifique de niveau international en recherche fondamentale, translationnelle et clinique. les créations du centre de recherche en cancérologie de lyon (crcl) aeres a+ sur le pôle lyon est et de la fondation synergie lyon cancer (slc) rtrs témoignent de cette dynamique. lyon recherche intégrée en cancérologie (lyric) est le site de recherche intégrée (siric) sur le cancer de lyon. lyric est porté par le centre léon bérard (clb) et les hospices civils de lyon (hcl), via le gcs lyon cancérologie universitaire (lcu). associé à l'inserm, le cnrs, l’inria, le crcl, la fondation slc, l’université lyon 1, ainsi que le centre international de recherche sur le cancer (circ), le site regroupe plus de 600 chercheurs en recherche fondamentale, translationnelle et clinique. lyric est piloté par un oncologue médical, le pr jy blay président de l'eortc, ainsi que deux directeurs adjoints le pr. a. puisieux directeur du crcl et le pr. jy scoazec, chef du service d'anatomie pathologique des hcl. lyric rassemble des équipes de recherche fondamentale et de transfert a+, ainsi que des équipes cliniques, toutes dotées d'une visibilité internationale. lyric dispose d’un programme de recherche fondamental bâti autour du crcl, et vise à faciliter le transfert de connaissances de la recherche fondamentale vers la recherche clinique et réciproquement, les objectifs seront de changer les paradigmes en biologie et en thérapeutique du cancer, d’identifier de nouvelles cibles thérapeutiques et des moyens de contourner la résistance à ces traitements. en effet, en 2011, les thérapies ciblées ont un nombre limité de cibles disponibles, et induisent l’émergence de résistance secondaire à terme. lyric met ainsi en place des programmes s'appuyant sur les expertises du crcl notamment déclinés vers la recherche translationnelle et la recherche clinique (sur les tumeurs modèles sur notre site, adénocarcinome du sein, lymphome, sarcome, net, tumeurs pédiatriques). les programmes de recherche intégrés de lyric explorent ainsi de nouvelles cibles modèle du crcl contribuant à la progression et à l’échappement tumoral : les voies de signalisation du développement embryonnaire dans les cellules tumorales (programme 1), les mécanismes de la tolérogénèse suscités par la réaction inflammatoire dans le stroma immunologique stroma (programme 2). le programme 3 explore des méthodologies innovantes pour cibler la destruction physique de la tumeur macroscopique, primaire ou métastatique. ces 3 programmes disposent d’objectifs individuels avec livrables et comprennent des projets spécifiques émergents. ils partagent aussi des approches transversales (profilage moléculaire prospectif des patients, des programmes innovants de recherche clinique internationaux). la recherche en shs et la collaboration avec le circ sur des projets d'épidémiologie moléculaire et la nutrition sont intégrés dans ces 3 programmes. les interactions avec les entreprises pharmaceutiques, les sociétés de biotechnologies et de technologies médicales sont intégrées au plan d'actions de lyric. la communication vers les professionnels de la santé, les réseaux régionaux, nationaux et internationaux sont des objectifs clés de lyric. plusieurs associations de patients de rayonnement national et national sont associées à lyric pour la diffusion des connaissances vers les patients et le grand public. l'objectif général de lyric à 5 ans est donc le développement de thérapeutiques personnalisées, adaptée à la cellule tumorale, au stroma immunologique et ciblant plus efficacement sur la tumeur macroscopique. lyric repose sur un réseau d'excellence associant une recherche intégrée sur le cancer et le développement thérapeutique, et sur une équipe multidisciplinaire d'experts internationaux. la labellisation de lyric comme l'un des siric français permettra de renforcer le positionnement international de lyon comme un site d’excellence de recherche en cancérologie de niveau mondial.

the site of lyon gathers a network of institutions dedicated to cancer research with a high level scientific visibility in basic, translational, and clinical research, as exemplified by the creations of the aeres a+ centre de recherche en cancérologie in lyon est (crcl), and of the synergy lyon cancer foundation (slc) cancer from the selective rtrs national call. the lyric, lyon integrated research institute in cancer, constitutes the integrated site for cancer research in lyon. it matches the objectives of a siric. lyric is co-piloted by the centre leon berard and hospices civils de lyon, through the lyon cancerologie universitaire structure. associated with inserm & cnrs, the crcl, the slc foundation, the university of lyon, the international agency for research in cancer (iarc), the site gathers >600 researchers in basic, translational and clinical research. the lyric is managed by a medical oncologist, prof jy blay, m.d. phd president of the eortc, together with two deputy directors, prof. a. puisieux, director of crcl, and prof jy scoazec, head of biopathology departments of hcl. lyric gathers a number of aeres a+ teams in basic, translational and clinical cancer research, in lyon, all of them with an international visibility. built around the crcl, lyric aims to streamline the transfer from the basic research laboratories to the clinical research area, and backwards to explore clinical observations using the tools and the paradigms of the basic research team for an accelerated qualitative scientific leap to the benefits of patient care. indeed, targeted oncogene therapies in 2011 have a limited set of available targets, and are doomed with the emergence of secondary resistance at midterm points. the lyric aims to provide innovative strategies to overcome these limitations, building on the expertises available at the crcl (reactivation of embryonic pathways & escape to immunosurveillance during tumor progression), in translational research and in clinical research (on breast carcinoma, lymphoma, sarcoma, net, pediatric tumors). the integrated research programs of lyric explore novel targets contributing to escape mechanisms. these are targets from the embryonic development pathways in tumour cells (program 1), and tolerogenic mechanisms reactivated by inflammatory processes within the immune stroma (program 2). program 3 explores novel methodologies for a safer and improved targeting and destruction of the macroscopic tumour. these 3 programs have specific goals, and specific emerging projects, but integrate also transversal approaches (prospective molecular profiling of all patients, innovative clinical research in national and international trials) and ultimately will merge toward common therapeutic research programs. research in human and social sciences and collaboration with iarc for joint projects on molecular epidemiology and nutrition are imbedded in these 3 programs. interactions with pharmaceutical company partners, biotech and medtech companies for drug & therapeutic development are integrated in lyric action plan. communication and dissemination toward the health care providers, general practitionners, regional, national and international networks is another major aim of lyric. several patient advocacy groups, with a national and international audience, are associated to lyric as actor of the dissemination program toward patients and the lay persons. the general aim of the lyric at year 5 is therapeutic development with an improved personalisation of the treatment, on the tumour cell, on the immune stroma, and on the macroscopic tumour, with paradigm changes in the biology and therapeutic of cancer. lyric is therefore a dedicated site of excellence for integrated cancer research and therapeutic development gathering a multidisciplinary team of international experts present on the lyon site. the labellisation of the lyric will enable major quantitative and conceptual leaps to the benefits of patient care, international world class positioning as a site of excellence in cancer research and treatment.
